I don't think the tranny end is too bad to do if you have cooler lines right there. But if you got headers on top of that then yes.

Truthfully I don't think the tranny end will be any trouble because the cables are very flexible and should be easy to route where you want them.

I had no problem with my Lokar set up. Same deal modified for a OE Carter set up and also moved onto a 1850 Holley.
Headers were not an issue with the Lokar cables at all. Routing was supper simple.
